Cellular regulation and molecular genetics of neurotransmitter transporters and receptors

Are you interested in molecular and cellular neurobiology? Do you want to understand how neurons control activity and availability of neurotransmitter receptors and transporters in the synapse? Role of PKMζ in the subiculum

Joint project between the labs of Jean-Francois Perrier (DNP) and Shohreh Issazadeh-Navikas (BRIC)
Rationale of the project
Long-term potentiation (LTP) is a form of plasticity that occurs in the limbic system. It is now well admitted that LTP is the cellular basis for learning and memory and scientists who contributed to its discovery were awarded the Brain Prize few weeks ago. The molecular mechanisms underlying the establishment of LTP are now well described. However, understanding how memory can be maintained over long periods (days to years) is a remaining challenge. It was recently suggested that Protein kinase Mζ (PKMζ), an atypical protein kinase C (PKC) isoform, plays a key role in the maintenance LTP. In addition, it was found in Alzheimer disease models that PKMζ accumulates in neurofibrillary tangles in brain areas involved in memory formation. Neurofibrillary tangles accumulate particularly in the hippocampal formation area called subiculum. Mice lacking IFN-β develop cognitive dysfunction and pathology in subiculum (Ejlerskov et. al. 2015 Cell). For that reason, our laboratories have decided to investigate the role PKMζ in the subiculum of normal versus mice lacking IFN-β and its receptor.
We are looking for one or two highly motivated master students to perform experiments in a slice preparation from the mouse brain as well as primary hippocampal neuronal cultures. We wish also to investigate if PKMζ is influences as a result of the genetic defects using Western blot and PCR techniques.
By performing patch-clamp recording of subicular neurons and calcium imaging with multi-photon microscopy, you will determine if and how PKMζ participate to memory formation in the subiculum in a healthy mouse and in the neurodegenerative models with dementia.
